Oxidative enzymes and phosphorylation in hepatic mitochondria from riboflavin-deficient rats.

چکیده
It has been known for many years that a number of different nutritional deficiencies lower the efficiency of food utilization. For example, riboflavin-deficient rats consume 15 to ‘20% more calories than are required to maintain control animals at the same weight (1). The lowered efficiency of food utilization in riboflavin deficiency has been attributed in part to incomplete oxidation of nutrients (2, 3). However, since flavoproteins are involved in the electron transport chain, it seemed possible that riboflavin deficiency and consequent decrease in flavin enzyme activities might affect the efficiency of oxidative phosphorylation. This possibility was tested with liver mitochondria from riboflavin-deficient rats. Highly significant alterations in the activity of certain oxidative flavin enzymes, and in the efficiency of oxidative phosphorylation of riboflavin-deficient mitochondria were found. In addition, some indications of changes both in the gross mass and character of the liver mitochondria have been observed.

Weanling rats were fed a simple riboflavin-deficient diet or the same diet supplemented with galactoflavin. Oxidative phosphorylation was studied in isolated hepatic mitochondria. Throughout the course of the experimental diets, a total of 8 weeks, there were no alterations in the respiratory control ratios or in the ADP/O ratios of the isolated mitochondria. Succinate dehydrogenase activity de...

Mice which had been on a riboflavin-free diet for 6-8 wk were given daily intraperitoneal injections of riboflavin. The hepatic mitochondria, which in the deficient animals were greatly enlarged, were restored to normal dimensions within 3 days. Normalization of the mitochondrial population was brought about by division of the giant organelles. Dividing mitochondria were characterized by a memb...
متن کاملApplication of Riboflavin-Deficient Rat Erythrocytes in the in Vitro Assay of Faba Bean Toxicity
Background: Faba bean toxicity (favism) is a common condition observed in our region. Objective: To develop a short and simple technique involving the use of riboflavin-deficient rats to test in vivo faba bean toxicity. Methods/Results: Sprague Dawley rats were maintained on a riboflavin-deficient diet and their vitamin B2 status was monitored by the assay of erythrocyte glutathione reductase (...
متن کاملThe effects of fenvalerate on hepatic and cerebral xenobiotic metabolizing enzymes in selenium and/or iodine deficient rats
Objective(s): Particularly in developing countries, selenium and/or iodine deficiencies are encountered and use of pesticides in agriculture are not well-controlled. Fenvalerate is a pyrethroid insectide used in agriculture and has applications against a wide range of pests. This study was designed to evaluate the effects of fenvalerate on hepatic and cerebral xenobiotic metabolizing enzyme act...
